Storage, cold chain and what actually degrades a lyophilised peptide
Freeze dried peptides are far more robust in transit than most resellers assume, and far more fragile once reconstituted. The difference matters for what you promise your customers.
There is a persistent belief in this trade that peptides are delicate things that expire the moment they leave a freezer. It is half right, and the half that is wrong costs resellers money on cold shipping they do not need while they store opened stock badly.
Dry powder is tough
A lyophilised peptide is a freeze dried solid with most of the water removed. Chemistry that would degrade it, hydrolysis of the peptide bond, oxidation of methionine and cysteine residues, deamidation of asparagine and glutamine, all need water and mobility to proceed at any speed. Take the water out and the reactions largely stop.
In practice this means a sealed vial of dry powder tolerates weeks at ambient temperature with no measurable change. That is why manufacturers ship internationally without cold packs and why an air freight leg sitting on a warm tarmac in Guangzhou is not the disaster it sounds like. If a supplier is charging you for cold chain on lyophilised product, ask what it is for.
Long term storage is where temperature earns its keep. Sealed, dry and at minus 20 degrees, two to three years is the normal expectation for most sequences. At 4 degrees, a year or more. At room temperature, think in months, and fewer of them in a humid climate.
Moisture is the actual enemy
Freeze dried peptide is hygroscopic. It pulls water out of the air, and the water it pulls in restarts exactly the chemistry the drying stopped.
The specific failure mode that catches people out is condensation. Take a vial from the freezer, open it immediately, and moist room air condenses on cold glass and cold powder. You have just added water to the one thing you were protecting from water. Let vials come to room temperature fully sealed before you open them, every time. This is the single most useful handling rule in the whole subject and it costs nothing.
Karl Fischer water content on a certificate is therefore a better predictor of how long a batch will last than the purity figure everyone fixates on. Under five percent is unremarkable. Over ten and the material has been dried poorly or stored open.
Freeze thaw cycling
Each freeze thaw cycle is a small opportunity for damage: local concentration changes, pH shifts as buffer components crystallise at different rates, and physical stress on the structure. One or two cycles is nothing. Twenty is a problem.
The practical answer for a reseller holding stock is to not use the working freezer as the storage freezer. Keep the bulk somewhere it will not be opened daily, and pull forward what you need for the week.
Reconstituted material is the fragile one
Once a peptide is in solution, everything above reverses. Water is now present in enormous excess, the molecule is mobile, and degradation proceeds at a rate you can measure in days.
Refrigerated at 4 degrees, most reconstituted peptides hold for two to four weeks depending on sequence, solvent and whether the solvent was bacteriostatic. At room temperature, assume hours to a couple of days. Light and oxygen both accelerate it, so amber glass and minimal headspace help.
This is a real distinction to communicate to your customers, because a customer who assumes the dry vial's shelf life applies to the reconstituted one will conclude the product was bad when it was the handling.
What this means for a reseller
- Do not pay for cold shipping on lyophilised product
- Store sealed at minus 20 for anything you are holding more than a few months
- Bring vials to room temperature before opening, without exception
- Split bulk from working stock so the bulk is not cycled
- Tell your customers plainly that dry and reconstituted shelf lives are different numbers
- Treat water content on the certificate as a shelf life signal, not a footnote
